LaPaglia was born in Adelaide, South Australia, the son of Maria Johannes (née Brendel), a secretary, and Gedio "Eddie" LaPaglia, an auto mechanic and car dealer. LaPaglia's mother was Dutch and his Italian father immigrated from Bovalino, Calabria at the age of eighteen. He is the younger brother of Australian actor Anthony LaPaglia. His other brother, Michael, is a car wholesaler in Los Angeles.
LaPaglia graduated from Rostrevor College, then the University of Adelaide with a degree in medicine. He worked three years as an emergency-room physician in Adelaide, Sydney and London. Feeling restricted, he decided to follow his brother into acting. In 1994, he moved to New York City where he joined the Circle in the Square Theatre School. He got his first break in 1996, when he joined the cast of TV show New York Undercover.
From 1998 until 2001 he played the role of Lt. Frank B. Parker on the science fiction series Seven Days. Afterwards, he played Detective Kevin Debreno, on The District from 2001-2004. He co-starred in the 2007 made for TV movie, Gryphon.
He's now living in Santa Monica, California with his wife, actress Ursula Brooks and their baby daughter.
Recent appearances include the final season of The Sopranos as himself, acting in the fictional Mafia-funded slasher film Cleaver.
LaPaglia has also appeared in episodes of NCIS, the first being aired in January 2008 called "Tribes", as FBI Special Agent Brent Langer. LaPaglia was set to return to the role in its sixth season as a recurring character, however in his next appearance in the sixth season premiere "Last Man Standing", he is suspected as a spy within the NCIS and shot by Agent Lee.
He appeared in the May 2008 issue of Car Craft magazine with his custom 6.1L Hemi '73 Dodge Challenger.
He had a recurring role in Cold Case in 2008-09.
In November, 2009 he appeared in an episode of the ABC television series, Castle, titled "Love Me Dead."
He features in the TV adaptation of Christos Tsiolkas' novel The Slap produced by Matchbox Pictures for ABC TV Australia, Screen Australia, Film Victoria and DCD Rights. It will be broadcast on BBC4 in October 2011.
